What are the minimum and maximum number of samples I can pool into a single tube?

How many samples can I pool in one prep?

The minimum number of samples to be processed and pooled at a time is 8 i.e., one column of the dried-in i1 Primer plate. The maximum number of samples to process and pool at a time depends on the input. Please see the following table:

How do I pool more than 48 samples?

The table below shows the volumes of purification reagents (PB and PS) to add to pooled samples after the THOR reaction.

When you intend to pool >48 samples, please prepare 2x 48 reaction pools and combine the two pools during the final purification at Step 11.
