WORKING COPY - Can I multiplex both UDI Sets A1 and B1 together?

Libraries prepared with UDI Set A1 and UDI Set B1 can be combined in the same sequencing lane with the following considerations:

Key differences

• i7 index sequences: Identical between A1 and B1

• i5 index sequences: Reverse complements of each other

Workflow design

• UDI Set A: Optimized for forward strand workflows

• UDI Set B: Optimized for reverse complement workflows

⚠️ Important notes

• Combining UDI Set A1 and UDI Set B1 results in a combinatorial indexing configuration rather than a true UDI setup; therefore, the full error-correction benefits of unique dual indexing will be lost.

• For best results, it is recommended to use UDI Sets A1–A4 or UDI Set B1 separately
