Which Sequencing Platforms are Lexogen Libraries Compatible with?

Full-length, indexed Lexogen libraries feature P5/P7 surface-binding adapter and internal sequencing primer-binding sequences that are directly compatible with all Illumina sequencing instruments and chemistries.

For adapter details please consult the “Sequencing” Appendix of the User Guide for the respective Lexogen library preparation kit.

Lexogen library compatibility with a range of popular and emerging sequencing platform technologies and instruments, is outlined below.

Library sequencing compatibility or conversion requirements outlined below may vary according to instrument, sequencing chemistry, or software version updates, or specific project requirements. Please contact the sequencing platform manufacturer for specific support regarding, library conversion protocol and product requirements, protocol performance outcomes, and sequencing run reagent and setting details.

Lexogen provides support only to ensure that libraries produced using our kits and protocols meet input requirements for sequencing provider workflows (e.g. input amounts, and input material type). Lexogen Technical Support is also happy to help you consult with sequencing provider support teams in order to verify compatibility for new conversion approaches. Lexogen’s support is based only on the verified experience of Lexogen, feedback from Lexogen customers, or method details from peer-reviewed publications.

Short-read platforms that require library conversion

The following platforms support sequencing after library conversion, for the following library types: QuantSeq (FWD / FFPE / REV / -Pool and combined with SLAMseq); CORALL (RTM / RTL / FFPE); Luthor (HD / HD-Pool); miRVEL (Discovery / Profiling), and Small RNA-Seq library types.

NOTE: Some conversion methods can also be performed on pools of indexed libraries (i.e. lanemixes).

Amplified full-length, indexed dsDNA libraries (or pools of libraries) are circularized to produce circular ssDNA library fragments.

Amplified full-length, indexed dsDNA libraries (or pools of libraries) have terminal Element-Biosciences adapters added, either through; circularization or PCR

Amplified full-length, indexed dsDNA libraries are subject to PCR to add G4-compatible flow cell-binding adapters (and indexes). Conversion cannot be performed on pools of indexed libraries (i.e. lanemixes).

Non-amplified cDNA, or amplified, full-length indexed dsDNA libraries are subject to PCR to add UG100-compatible adapters (with or without indexes).

Amplified full-length, indexed dsDNA libraries are subject to PCR to add PacBio compatible adapter sequences. Conversion is not recommended for pools of indexed libraries (i.e. lanemixes).

Long-read sequencing platforms

Long-Read Sequencing Platform technologies are compatible with Lexogen’s TeloPrime Full-Length cDNA Amplification Kit (Cat. No. 013).

IMPORTANT: Conversion requirements for new sequencing instruments, and/or new vs legacy versions of kit chemistry, should be confirmed with the instrument manufacturer directly.

Methods

  • Custom forward and reverse primers featuring 5' overhangs that encode PacBio or ONT adapter sequences are required.

  • These custom primers replace the FP and RP PCR primers for TeloPrime cDNA Amplification.

  • Using custom primers with adapter sequences during TeloPrime reverse transcription is also possible.

  • The amplified TeloPrime cDNA can then be used for downstream PCR or adapter-ligation based library construction.

  • To confirm the specific method and kit requirements please consult with PacBio or ONT technical support.
