What is the difference between the Lexogen i5 6 nt Dual Indexing Add-on Kits and the Lexogen UDI 12 nt Unique Dual Indexing Kits?
The Lexogen i5 6 nt Dual Indexing Add-on Kits (Cat. No. 047) contain the indexed i5 adapters and PCR buffer (Dual PCR Mix) for dual indexing. They can be used to introduce dual indexing for all Lexogen library prep kits that include the Lexogen i7 6 nt Indices (Cat. No. 015, 016, 095, 146, and 147). The indices of the i7 and i5 6 nt Index Sets are 6 nt long.
The Lexogen UDI 12 nt Unique Dual Indexing Sets contain pre-mixed i5 and i7 adapters, each including 12 nt long indices. Lexogen 12 nt UDI patented design optimizes the Levenshtein distance between each index. This allows rescuing a significant number of reads that would otherwise be lost because of index sequence errors occurring during the sequencing step.

The Lexogen 12 nt Unique Dual Indexing Index V2 Add-on Kits additionally include the PCR Enzyme Mix (PE) and PCR Mix (PM) required for dual indexing PCR. In the previous version of the UDI Add-on kits (V1), the Enzyme Mix (E) and the Dual PCR Mix (Dual PCR) were provided instead of PE and PM.
When using the 12 nt UDI Sets for the introduction of Unique Dual Indexing to libraries prepared with Lexogen kits that include the Lexogen i7 6 nt Indices (Cat. No. 015, 016, 095, 146 and 147), the PCR Mix (PCR, yellow) and the Enzyme Mix 3 (E3) from the library prep kits have to be replaced with the PCR Mix (PM) and the PCR Enzyme Mix (PE) from the UDI 12 nt Add-on kits V2 (or with E and Dual PCR when using the V1 of the kits), and the 12 nt UDIs must be used instead of the i7 6 nt indices.
For convenience, the QuantSeq FWD and CORALL kits are available as bundles containing the UDI 12 nt Sets instead of the i7 6 nt Index Sets (Cat. No. 191 – 196 for QuantSeq FWD, 171 – 176 and 183 - 186 for CORALL Total and 177 – 182 for CORALL mRNA).
