Can I use the enzyme from the PCR Add-on kit to perform QuantSeq-Pool Endpoint PCR?

Yes. The PCR Add-on and Reamplification Kit V2 for Illumina (Cat. No. 208) can be used to perform both the qPCR assay and the Endpoint PCR on QuantSeq-Pool libraries. Both kits use the same PCR system, which includes both the PCR Mix “PM” and the PCR enzyme Mix “PE”. ATTENTION: The “P7” , “P5” and “RE” primers of the PCR Add-on and Reamplification Kit V2 for Illumina (Cat. No. 208) are not indexed. Therefore if you want to multiplex more than 96 samples, UDIs must be used during the Endpoint PCR, instead of the generic primers “RE”, “P7” and “P5”.
