How do I choose the number of PCR cycles to use for library amplification ?

Cycle numbers for endpoint PCR are indicated in the table in Appendix B of the respective miRVEL Discovery or miRVEL Profiling Small RNA-Seq Library Prep Kit User Guides.

ATTENTION: These ranges are provided as a guideline only for choosing an approximate number of cycles for the Endpoint PCR Test. For example, we recommend performing 16-17 PCR cycles when using an input amount of 100 ng total RNA. Depending on the small RNA content of the initial RNA sample, an additional 1 or 2 cycles could be added.

NOTE: When using the miRVEL Profiling kit, we recommend performing a qPCR assay to determine the optimal number of PCR cycles to use for the endpoint PCR. For more information, please see the following FAQ: Do I need to perform a qPCR assay to determine the number of PCR cycles for the EP PCR?
